Just use the ini file. There's an example in PortableThunderbird\Other\PortableThunderbirdSource. Copy the PortableThunderbird.ini file to the PortableThunderbird directory... next to PortableThunderbird.exe. Change the AdditionalParameters line to be:
You can also it as a parameter to PortableThunderbird.exe and it'll just it to thunderbird.exe.
